Seven in absentia homolog 3 (Siah-3), encoded by the SIAH3 gene in humans, functions as a negative regulator of PRKN translocation to damaged mitochondria. It acts by destabilizing PINK1 protein, thereby inhibiting PRKN targeting to dysfunctional depolarized mitochondria. Siah-3 is localized to the mitochondrion, where it plays a critical role in modulating mitochondrial quality control pathways, with implications for cellular stress responses and neurodegenerative disease mechanisms.
