This fragment represents a key regulatory component of the IKK complex, essential for NF-κB signaling pathway activation.

    NF-kappa-B essential modulator (NEMO), encoded by the IKBKG gene, serves as the regulatory subunit of the IκB kinase (IKK) core complex. It plays a critical role in phosphorylating inhibitors of NF-κB, leading to their degradation and subsequent activation of NF-κB transcription factors. NEMO recognizes and binds both 'Lys-63'-linked and linear polyubiquitin chains with high affinity, facilitating IKK activation through multiple signaling pathways. It is also involved in antiviral innate immune responses via TLR3 and IFIH1-mediated mechanisms and contributes to viral activation of IRF3. Subcellularly, NEMO localizes to the cytoplasm and nucleus, with sumoylated forms accumulating in the nucleus under genotoxic stress. This recombinant fragment (50-357) provides a stable, soluble unit for investigating NEMO's structural and functional properties in NF-κB signaling and immune regulation.
