Its sequence is Tyr-D-Leu-Phe-Ala-Asp-Val-Ala-Ser-Thr-Ile-Gly-Asp-Phe-Phe-His-Ser-Ile-NH2, featuring a D-leucine residue at position 2. This peptide belongs to the deltorphin subfamily, characterized by the presence of a D-amino acid in the second position, which is crucial for bioactivity in related peptides.

The natural [D-Leu2]deltorphin exhibits low affinity for δ-opioid binding sites in both peripheral tissues and the central nervous system. However, a shorter synthetic amidated analogue, [D-Leu2]deltorphin(1-10), demonstrates significant agonistic potency and selectivity for δ-opioid receptors, comparable to enkephalins.
